Semaglutide is a glucagon-like peptide-1 (GLP-1) receptor agonist with an EC50=6.2 pM in reporter gene experiments on BHK cells expressing the human receptor. In type II diabetes in db/db mice, semaglutide reduces blood glucose levels, stimulates insulin and inhibits glucagon secretion in a glucose-dependent manner. Semaglutide is a longer-acting alternative to Liraglutide (MZ7351) and is structurally similar to GLP-1. In the MPTP-induced Parkinson model, semaglutide (25 nmol/kg) prevented the reduction of dopaminergic neurons in the Semelin ganglion and increased lipid peroxidation in the Semelin ganglion and striatum. as well as improving the movement coordination of animals in roller experiments and footprint analysis. Semaglutide has anti-obesity activity. Possible mechanisms of action of semaglutide-induced weight loss include reduced energy intake, better dietary control, and a relatively lower preference for fat and caloric-heavy foods.
